Abstract:

Melatonin regulates some major physiological processes such as maturation and function of reproductive system, pubertal development, seasonal reproduction and adaptation. The activation of melatonin hormone is mediated by melatonin receptor. Previous studies showed melatonin receptor 1A gene (MTNR1A) is highly polymorphic in seasonally breeding species. The aim of the present study was detection of MTNR1Apolymorphism and its association with production (body weights at birth, 1, 3, 6, 9 and 12 month of age) and reproduction (litter size) traits in Zandi sheep using PCR-RFLP methodology. Hundred blood samples were randomly collected and genomic DNA was isolated using modified salting out method. A large fragment of exon 2 of MTNR1A gene was amplified by PCR using specific primer pairs. After amplification, the PCR product was digested with MnII endonuclease. Restricted digestion allowed the determination of two alleles (M, m) and two genotypes (MM, Mm) with frequencies of 0.91, 0.09 and 0.82, 0.18, respectively. Least square means showed that MM individuals had higher body weight at one month of age (BW1) than Mm individuals (P

Polymorphism of the Melatonin Receptor 1A Gene and Its Association with Litter Size in Zel and Naeini Sheep Breeds

The influence of melatonin receptor 1A gene on litter size was studied in 150 ewes from Zel (n=100) and Naeini (n=50) sheep breeds. Two restriction fragment length polymorphism (RFLP) analyses were done to determine all genotypes occurring by variations at MnlI and RsaI restriction sites. The M and C were the most frequent alleles in both of these breeds. The MM genotype was predominant in Zel ...

Study of Dopamine Receptor Gene Polymorphisms and Their Association with Growth and Egg Production Traits in West Azerbaijan Native Chicken

The objective of this study was to search for single nucleotide polymorphism (SNP)-type polymorphisms in the dopamine D1 receptor in West Azerbaijani native chicken and look for their association with egg production and body weight traits of chickens by polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP). For this purpose 180 blood samples were taken from nativ...

Association between Melatonin Receptor 1A (MTNR1A) Gene Polymorphism at the MnlI Site and Production Traits in Shal and Crossbreeding between Shal and Romanov

The objective of this study was to evaluate the relationship between polymorphism at the MnlI site of MTNR1A gene and production traits of Shal and Romanov crossbreeding. The crossbreeding between Shal and Romanov was done on 600 Shal ewes. For this reason, Shal ewes were laparoscopically and artificially inseminated with the semen of Romanov breed. For the preparation of genomic DNA, a total o...

Association of IGF-I Gene Polymorphisms with Carcass Traits in Iranian Mehraban Sheep Using SSCP Analysis

Molecular genetics selection on individual genes is a promising method to genetically improve economically important traits in livestock. The insulin like growth factor-I (IGF-I) gene may play important roles in growth of multiple tissues, including muscle cells, cartilage and bone. The objectives of the present study were the estimate the haplotype frequencies of the IGF-I gene polymorphisms i...
